I have a B52 AT100 amp and on the back it has a switch for different ohm selections. I can choose between 4, 8 or 16 ohm and I am currently running it on 4ohm since I only have one 4x12 cab. Can someone explain to me what would be the best setting to run it on and why. I just don't know a whole lot about ohm's. I was told the lower the ohm's the more power you are pushing.

By adding more speakers ( In paralell ) your giving the output more pathways for the current to travel through and so the current increases - Due to Ohms law - if the voltage is constant and the current increases - then the total resistance - (Ohms) must be lower.

You could measure the Resistance with a multimeter set on Ohms, but because the multimeter uses DC, and the speakers work on AC the reading will be slightly out.
ie. An 8 ohm speaker will give a reading of 7 ohms.

Failing that you could open up the cab and see if the speakers are marked, it might say ' x ' amount of ohms or a number with the Greek symbol Omega - which stands for Ohms.
Remember though that when speakers are wired in paralell the total resistance decreases

the two inputs on my cab tell me which to use for the different ohm settings. I am just wondering which setting will give me the best sound 4, 8, or 16 and which is best for the longevity of my amp.

ideally you match the Resistance or impedance( as it's known ) setting to the cab,

4x sixteen ohm speakers will give a combined resistance of 4 Ohms

2x sixteen ohm speakers will give a combined resistance of 8 ohms

Hope i'm helping
