When you play this song with just one guitar you can either play the lead guitar part during the whole song, or use a combination of the two. If you want to combine the two guitar parts you use the arpeggios, while the vocal phrase is happening. It's a good idea to cut it short a little early- for example the 2nd time you reach the b7, so you can get ready to play the lick. If it's too hard switching between the slide and the rhythm part you can always just use regular, fretted blues licks instead. Once you reach the final outro jam you stick to the lead part.
