Measure 1 (or bar 1) is simply a reference showing all 3 chords to be used in the example progression. The practice tune actually starts at measure 2.

They happen to be on the same page, so the notation transcriber just made them part of the same overall measure counting.

The turnaround is traditionally the last 2 measures of a 12 bar blues. So the turnaround will be both measures 11 & 12 of this blues (A - I chord & E - V chord). Or if you are following the numbers in red, that's measures 12 & 13.
